FX not effecting

Anyone know why a striing of FX applied to a clip ( say B+W/CONTRAST and BRIGHTNESS) won't show up on the preview window on replay ?

You've clicked the button on the preview for bypass FX. Click it again (it's the one with the black/white circle).
