For volume changes you need to add a volume envelope. Just select the audio track you want to control the volume on and press the V key. (Alternately, go to the menu and select Insert-->Audio Envelopes-->Volume but pressing V is so much faster) ;-) This will add a blue line across the audio track that is the envelope. Now double click on where toy want a volume change to take place and drag the line up to increase the volume or down to decrease it. Add as many points as you need to get the changes you want.

Effects (FX) work on entire events. So for effects you have to split the event so that you now have two or three events. To only split the audio you have to ungroup it from the video. Then you can group it back again so they all move together. Here’s what you do:

1. Select the event you want to add the FX to
2. Press the U key to ungroup the Video and Audio
3. Select the Audio only
4. Place the cursor where you want the FX to start and press the S key to split the event
5. Place the cursor where you want the FX to end and press the S key to split the event
6. (Optional) Ctril-Click on the four (4) events (1 Video and 3 Audio you just made) and press the G key to Group them back together so they move as one.
7. Now just select the audio event in the middle and apply the FX you want. Only that portion of the audio will be affected.

Hope this was clear enough to follow without pictures.
