With Photoshop or a similar application. Vegas allows you to drop the default photoshop file right on the timeline as-is. Use masking (in Photoshop) to make whatever part of the image you wish trasnparent, place on a track ABOVE your regular video track.

Are you aware that you can use the built in media generators simply as a starting point and then edit the generated text (with transparent backround for instance) in any way you wish?

Ocean's right. And it sure does most of the the jobs and is easy to use. Although I often use Photoshop 'cause I feel more like it; color shifting, hue, filters, effects...

With the built-in generator, how do you put the text on top of a button? The transparent background I know can be done.
