You pull down the middle button at the top of the clip.

Or the Level Slider if the clip is on a track by itself.

Or place a video or image with an Alpha Layer on a track above.

And/or apply a Track Compositing Effect if you want the layers to interact.

@mark-y Does the video have to be a special format or recorded in a special way to get that alpha layer? Could I re-encode it perhaps, to get that transparent quality?

Your overlay video or image must be created as RGBA32, not RGB24, --or-- a Mask or Chroma Key or Secondary CC can be used to reveal the transparency. Only certain formats support an Alpha Layer, and they can be found by using the forum search feature.
